Freezing of Gait in Parkinsonism and its Potential Drug Treatment
Authors: Li-Li Zhang, S. Duff Canning and Xiao-Ping WangFreezing of gait (FOG) is a heterogeneous symptom. Studies of treatment for FOG are scarce. Levodopa and monoamine oxidase inhibitors (rasagiline and selegiline) have shown effective improvement for FOG. Other drugs, such as L-threo-3, 4-dihydroxyphenylserine, amantadine, and botulinum toxin have exhibited some beneficial effects. The present review summarizes the potential drug treatment for FOG in Parkinsonism.

Drug Therapy for Behavioral and Psychological Symptoms of Dementia
Authors: Feng Wang, Ting-Yi Feng, Shilin Yang, Maurice Preter, Jiang-Ning Zhou and Xiao-Ping WangDementia, which can be induced by diverse factors, is a clinical syndrome characterized by the decline of cognitive function. Behavioral and psychological symptoms of dementia (BPSD) include depression, agitation, and aggression. Dementia causes a heavy burden on patients and their caregivers. Patients with BPSD should be assessed comprehensively by practitioners and offered appropriate non-pharmacologic and ph Read More

Current Therapy of Drugs in Amyotrophic Lateral Sclerosis
Authors: Haiyan Lu, Wei D. Le, Ya-Ying Xie and Xiao-Ping WangAmyotrophic lateral sclerosis (ALS), commonly termed as motor neuron disease (MND) in UK, is a chronically lethal disorder among the neurodegenerative diseases, meanwhile. ALS is basically irreversible and progressive deterioration of upper and lower motor neurons in the motor cortex, brain stem and medulla spinalis. Riluzole, used for the treatment of ALS, was demonstrated to slightly delay the initiation of respi Read More

Current Drug Managements of Wilson’s Disease: From West to East
Authors: Wen-Jie Li, Chen Chen, Zhi-Fei You, Ren-Min Yang and Xiao-Ping WangWilson’s disease (WD), also called hepatolenticular degeneration, is an autosomal recessive inheritance disorder of copper metabolism characterized by the multiple mutations in the ATP-ase 7B gene of chromosome 13q. About half of the WD patients have neurological or psychiatric symptoms. As WD is a kind of medicable or nearly curable neurodegenerative disease in the field of medicine, early consideration/exam Read More

Current Treatment Options for Alzheimer’s Disease and Parkinson’s Disease Dementia
Authors: Jennifer Y.Y. Szeto and Simon J.G. LewisAlzheimer’s disease (AD) and Parkinson’s disease (PD) are the two most common neurodegenerative disorders encountered in clinical practice. Whilst dementia has long been synonymous with AD, it is becoming more widely accepted as part of the clinical spectrum in PD (PDD). Neuropsychiatric complications, including psychosis, mood and anxiety disorders, and sleep disorders also frequently co-exist with cogni Read More

Current Pharmaceutical Treatments and Alternative Therapies of Parkinson’s Disease
Authors: Jie Dong, Yanhua Cui, Song Li and Weidong LeOver the decades, pharmaceutical treatments, particularly dopaminergic (DAergic) drugs have been considered as the main therapy against motor symptoms of Parkinson's disease (PD). It is proposed that DAergic drugs in combination with other medications, such as monoamine oxidase type B inhibitors, catechol-O-methyl transferase inhibitors, anticholinergics and other newly developed non-DAergic drugs can make a better co Read More

Revisiting the Medical Management of Parkinson’s Disease: Levodopa versus Dopamine Agonist
Authors: Jinglin Zhang and Louis Chew-Seng TanThe optimal treatment strategy for Parkinson’s disease has been debated for decades. The introduction of levodopa (LD) treatment is frequently delayed because of theoretical concerns about its toxicity or the risk of drug-induced motor complications. These concerns have resulted in “LD phobia” with clinicians selecting dopamine agonist (DA) over LD as initial therapy. More recently, a shift in the treatment approach towar Read More

AChE Inhibition-based Multi-target-directed Ligands, a Novel Pharmacological Approach for the Symptomatic and Disease-modifying Therapy of Alzheimer’s Disease
Authors: Yu Wang, Hao Wang and Hong-zhuan ChenAlzheimer's disease (AD) is the most common form of dementia in elder people, characterised by a progressive decline in memory as a result of an impairment of cholinergic neurotransmission. To date acetylcholinesterase inhibitors (AChEIs) have become the most prescribed drugs for the symptomatic treatment of mild to moderate AD. However, the traditional “one molecule-one target” paradigm is not sufficient and appr Read More

Drug Delivery Systems for Imaging and Therapy of Parkinson’s Disease
Authors: Mine Silindir Gunay, A. Yekta Ozer and Sylvie ChalonBackground: Although a variety of therapeutic approaches are available for the treatment of Parkinson’s disease, challenges limit effective therapy. Among these challenges are delivery of drugs through the blood brain barier to the target brain tissue and the side effects observed during long term administration of antiparkinsonian drugs. The use of drug delivery systems such as liposomes, niosomes, micelles, nanoparticles, n Read More

Molecular, Cellular and Clinical Aspects of Intracerebral Hemorrhage: Are the Enemies Within?
Authors: Cássia Righy, Marcelo T. Bozza, Marcus F. Oliveira and Fernando A. BozzaHemorrhagic stroke is a disease with high incidence and mortality rates. In addition to the mass lesions that result from hemorrhagic stroke, substances such as the blood-derived products (BDP) (hemoglobin (Hb), heme and iron) induce a potent inflammatory response and exert direct toxic effects on neurons, astrocytes, and microglia. In the present review, we discuss the mechanisms of brain injury secondary to hemo Read More
